it helps some times I just had to do the same exact type of thing...here is how you add a column.
code :
If Not IsPostBack Then <br>

        Dim field As New TemplateField
        field.HeaderText = "Name of Column"
        Dim col As DataControlField = field
        GridView.Columns.Add(col)

        End If
e.row.cells(cellnumber from 0 to N).controls.Add(data)
Dim Dbconn As SqlConnection
Dim Dbcmd As SqlCommand

Dbcmd = New Data.SqlClient.SqlCommand()
Dbcmd.Connection = Dbconn
Dbcmd.CommandType = Data.CommandType.Text

Dbcmd.Commandtext = "select * from table"
dbconn.open()

//then you need a data reader

dim dr as sqlclient.sqldatareader
dr = dbcmd.executereader
while dr.read
add each item to a list
end while

hope this fix your issue You've got your code right, but instead I've put it in the CellValueChanged event and check to make sure it was the same cell you're looking for. This will get fired if the cell's value changes, or if a new row is added and subsequently gets a value assigned.
code :
    private void dataGridView2_CellValueChanged(object sender, DataGridViewCellEventArgs e)
    {
        if (e.ColumnIndex == 5)
            textBox9.Text = CellSum().ToString();
    }

    private double CellSum()
    {
        double sum = 0;
        for (int i = 0; i < dataGridView2.Rows.Count; ++i)
        {
            double d = 0;
            Double.TryParse(dataGridView2.Rows[i].Cells[5].Value.ToString(), out d);
            sum += d;
        }
        return sum;
    }

this one helps. If you are sourcing your DataGridView from a DataTable, I've already created the following solution when I once encountered this issue.*
The idea is to indicate which columns require summing, which column and text will indicate the "Total" label, then take the underlying DataTable and manually sum the indicated columns. These sums and the label are used to create a new DataRow, which is added to the end of the table. When any changes are made - programmatic adding of a new row, deleting a row, sorting - the old sum row is removed and a new one is calculated then added.
